Are there any courses available which teach everything I could ever want to know about NSIS? How to use it, tricks of the trade, tips, best practices, etc.?

I don't think there are any courses. You generally just play around on your own (including RTFM) and ask a specific question if you run into trouble.
You can find some setup guidelines from MS but they often have a MSI focus and best practices have changed over the years.
